Potassium Channel Localization via Dendritic Voltage Clamp Recordings

چکیده
Here I propose to extend, analyze and apply a promising acceleration of the method of Schaefer et al. [Sch]. They focus on recovering the conductance and kinetics of potassium currents at the clamp site. Let us begin, as they do, with the simple case of a straight resting fiber of length ` and radius a with a single noninactivating potassium current. We specify a voltage command V (t) at the clamp site, x = xc, and measure the requisite applied current, I(t), at xc. The associated cable equation is

O3: Pharmacological Modulation of Thalamic KCNQ-Potassium Channels: Insight from Knock-out Mice
The channels belonging to the KCNQ gene family consist of 5 different subtypes, which assemble as pentameric channels. The KCNQ2-5 subunits are highly expressed in the ventrobasal thalamus (VB) where they function primarily as KCNQ2/3 heteromers. They underlie an outward potassium (K+)-current, called M-current (IM), which provides a hyperpolarizing drive, thus regulating neuronal excitability....
متن کاملPersistent sodium and calcium currents in rat hypoglossal motoneurons.
Voltage-dependent persistent inward currents are thought to make an important contribution to the input-output properties of alpha-motoneurons, influencing both the transfer of synaptic current to the soma and the effects of that current on repetitive discharge. Recent studies have paid particular attention to the contribution of L-type calcium channels, which are thought to be widely distribut...
متن کاملSynaptic integration and dendritic excitability in cerebellar Purkinje neurons
Dendrites receive most of the synaptic input to the neuron, but their contribution to synaptic integration is not well understood. In this thesis I investigate synaptic integration in Purkinje neurons in cerebellar slices, focusing on the contribution of dendritic excitability using simultaneous somatic and dendritic patch-clamp recordings. I first investigated the ionic conductances underlying...

To understand functions of a single neuron, such as propagation and generation of synaptic or action potentials, a detailed description of the kinetics and distribution of the underlying ionic conductances is essential. In voltage-clamp experiments, incomplete space clamp distorts the recorded currents, rendering accurate analysis impossible. Here, we present a simple numerical algorithm that c...
متن کاملPreparation of Drosophila central neurons for in situ patch clamping.
Short generation times and facile genetic techniques make the fruit fly Drosophila melanogaster an excellent genetic model in fundamental neuroscience research. Ion channels are the basis of all behavior since they mediate neuronal excitability. The first voltage gated ion channel cloned was the Drosophila voltage gated potassium channel Shaker(1,2). Toward understanding the role of ion channel...
